Presence of Toxoplasma gondii infection in brain as a potential cause of risky behavior: a report of 102 autopsy cases

Toxoplasmosis was linked to impairment in brain function, encompassing a wide range of behavioral and neuropsychiatric changes. Currently, the precise localization of Toxoplasma gondii in the human brain is limited and the parasite DNA was not found in population-based screening of autopsy cases. The aim of proposed study was to identify the presence of parasite DNA within the brain and its association with risky behavior and alcohol consumption in postmortem examination. Preliminarily, 102 cases with certain circumstances of death at time of forensic autopsy was included. Due to high risk of bias, the females were excluded from the analysis and final study group consists 97 cases divided into three groups: risky behavior, inconclusively risky behavior, and control group. The obtained tissue samples for Nested PCR covered four regions of the brain: symmetric left/right and anterior/posterior horns of lateral ventricles comprising lining ependyma and hippocampus. The second type of material comprised blood evaluated for antibodies prevalence using ELISA and alcohol concentration using HS-GC-FID. Analysis demonstrated 16.5% prevalence concerning the parasite DNA presence in examined brain tissue samples without specific distribution and association with age at death or days after death until an autopsy was performed. Results have shown correlation between occurrence of risky behavior leading to death and higher proportions of positive parasite DNA presence within the brain. Correlation was not observed between parasite DNA presence and excessive alcohol consumption. Conducted screening demonstrated correlation between parasite DNA presence in the brain with risky behavior and provided new information on possible effects of latent toxoplasmosis.

Toxoplasma gondii is a successful protozoan parasite that cycles between definitive fetid hosts and a broad range of intermediate hosts, including rodents and humans. Within intermediate hosts, this obligate intracellular parasite invades the small intestine, inducing an inflammatory response. Toxoplasma infects infiltrating immune cells, using them to spread systemically and reach tissues amenable to chronic infection. An intact immune system is necessary to control life-long chronic infection. Chronic infection is characterized by formation of parasite cysts, which are necessary for survival through the gastrointestinal tract of the next host. Thus, Toxoplasma must evade sterilizing immunity, but still rely on the host's immune response for survival and transmission. To do this, Toxoplasma exploits a central cost-benefit tradeoff in immunity: the need to escalate inflammation for pathogen clearance vs. the need to limit inflammation-induced bystander damage. What are the consequences of sustained inflammation on host biology? Many studies have focused on aspects of the immune response that directly target Toxoplasma growth and survival, commonly referred to as "resistance mechanisms." However, it is becoming clear that a parallel arm of the immune response has evolved to mitigate damage caused by the parasite directly (for example, egress-induced cell death) or bystander damage due to the inflammatory response (for example, reactive nitrogen species, degranulation). These so-called "disease tolerance" mechanisms promote tissue function and host survival without directly targeting the pathogen. Here we review changes to host metabolism, tissue structure, and immune function that point to disease tolerance mechanisms during Toxoplasma infection. Toxoplasmosis is a parasitic disease caused by Toxoplasma gondii with a globally widespread distribution. The aim of this systematic review and meta-analysis was to characterize the association between T. gondii infection and multiple sclerosis (MS). The data were systematically collected from the English electronic databases up to April 2017. The research process resulted in the identification of five studies related to the subject of interest entailing 669 MS patients and 770 controls. The pooled prevalence rates of T. gondii infection in the MS patients and controls were estimated as 32.4% (95% CI: 27.4-38.6) and 39.1% (95% CI: 29.1-50.5), respectively. By random effect model, the combined odds ratio was 0.72 (95% CI: 0.49-1.06) with P = .0961. Although this meta-analysis study showed a lower seroprevalence of T. gondii in the MS patients as compared with that in the control group, no significant association was found between toxoplasmosis and MS disease. Further investigations are recommended to determine the detailed association between MS patients and T. gondii infection.

The observation that certain species of parasite my adaptively manipulate its host behaviour is a fascinating phenomenon. As a result, the recently established field of 'host manipulation' has seen rapid expansion over the past few decades with public and scientific interest steadily increasing. However, progress appears to falter when researchers ask how parasites manipulate behaviour, rather than why. A vast majority of the published literature investigating the mechanistic basis underlying behavioural manipulation fails to connect the establishment of the parasite with the reported physiological changes in its host. This has left researchers unable to empirically distinguish/identify adaptive physiological changes enforced by the parasites from pathological side effects of infection, resulting in scientists relying on narratives to explain results, rather than empirical evidence. By contrasting correlative mechanistic evidence for host manipulation against rare cases of causative evidence and drawing from the advanced understanding of physiological systems from other disciplines it is clear we are often skipping over a crucial step in host-manipulation: the production, potential storage, and release of molecules (manipulation factors) that must create the observed physiological changes in hosts if they are adaptive. Identifying these manipulation factors, via associating gene expression shifts in the parasite with behavioural changes in the host and following their effects will provide researchers with a bottom-up approach to unraveling the mechanisms of behavioural manipulation and by extension behaviour itself.

Significance of chronic toxoplasmosis in epidemiology of road traffic accidents in Russian Federation

Studies carried out in Moscow residents have revealed that the prevalence of chronic toxoplasmosis is very close to those in countries of Eastern and Central Europe. Our findings also demonstrated a statistically significant relationship between the rate of traffic accidents and the seroprevalence of chronic toxoplasmosis in drivers who were held responsible for accidents. The latter was 2.37 times higher in drivers who were involved in road accidents compared with control groups. These results suggest that the consequences of chronic toxoplasmosis (particularly a slower reaction time and decreased concentration) might contribute to the peculiarities of the epidemiology of road traffic accidents in the Russian Federation and might interfere with the successful implementation of the Federal Programme named "Increase road traffic safety" . Suggestions for how to address overcome this problem are discussed in this paper.

Purpose of review The apicomplexan protozoan Toxoplasma gondii has a striking predilection for infecting the central nervous system and has been suggested as a risk factor for schizophrenia. Here, we address some of the mechanisms by which T. gondii achieves this by manipulating signaling pathways of the host brain cells. Recent findings Recent years have brought notable progress in the understanding of how the opportunistic parasite T. gondii establishes a successful infection in mammalian brain by secreting effector molecules that manipulate multiple cell functions. Many pathways involved in this inter-kingdom signaling, such as dopaminergic, GABAergic and kynurenine pathways, also have key roles in the development of schizophrenia. More understanding of T. gondii-brain cell interaction holds the key to unlocking the mystery of T. gondii-mediated schizophrenia pathogenesis. Summary T. gondii usurps a variety of host signaling pathways to ensure physiological adaptation, evasion of host immune defense systems, and efficient replication. A detailed knowledge of T. gondii signaling molecules involved in this cross-kingdom communication with host brain cells will probably provide novel means of pharmacologically manipulating host cellular pathways to promote efficient elimination of the parasite and may permit the development of new schizophrenia-modifying therapeutics.

Recently, many studies have shown that latent Toxoplasma infections could have serious impacts on human health. Here we ran a cross-sectional study in a population of 1486 volunteers. The results showed that 333 infected subjects scored worse than 1153 controls in 28 of 29 health-related variables. Similarly, they reported higher rates of 77 of a list of 134 disorders reported by at least 10 participants of the study. Toxoplasmosis was associated most strongly with musculoskel- etal ( τ = 0·107, P < 0·0005), followed by neurological ( τ = 0·088, P < 0·0005), immune ( τ = 0·085, p < 0·0005), metabolic ( τ = 0·079, P < 0·0005), respiratory ( τ = 0·068, P = 0·0001), allergic ( τ = 0·053, P = 0·004), digestive system ( τ = 0·052, P = 0·004) and mental health disorders ( τ = 0·050, P = 0·008). Results of the present cohort study, along with the previous data from many case-control studies or ecological studies suggest that latent toxoplasmosis represents a large and so far underrated public health problem

Cat scratches, not bites, are associated with unipolar depression – cross-sectional study

Background: A recent study performed on 1.3 million patients showed a strong association between being bitten by a cat and probability of being diagnosed with depression. Authors suggested that infection with cat parasite Toxoplasma could be the reason for this association. Method: A cross sectional internet study on a non-clinical population of 5,535 subjects was undertaken. Results: The subjects that reported having been bitten by a dog and a cat or scratched by a cat have higher Beck depression score. They were more likely to have visited psychiatrists, psychotherapists and neurologists in past two years, to have been previously diagnosed with depression (but not with bipolar disorder). Multivariate analysis of models with cat biting, cat scratching, toxoplasmosis, the number of cats at home, and the age of subjects as independent variables showed that only cat scr atching had positive effect on depression ( p = 0.004). Cat biting and toxoplasmosis had no effect on the depression, and the number of cats at home had a negative effect on depression ( p = 0.021). Conclusions: Absence of association between toxoplasmosis and depression and five times stronger association of depression with cat scratching than with cat biting suggests that the pathogen responsible for mood disorders in animals-injured subjects is probably not the protozoon Toxoplasma gondii but another organism; possibly the agent of cat-scratched disease – the bacteria Bartonella henselae .

About 30–50% of the world human population are infected with the protozoan parasite Toxoplasma gondii ( Nicolle et Manceaux, 1908). Latent toxoplasmosis has many specific behavioural and physiological effects on the human body and influences the course of pregnancy, including secondary sex ratio of children of infected mothers. It was suggested that an increased concentration of glucose could be the proximate cause of increased sex ratio. There are some indirect indications of possible association between toxoplasmosis and certain forms of diabetes. Here we searched for a possible link between latent toxoplasmosis and the level of glu - cose in the blood. In a cross-sectional study, we found that pregnant women with latent toxoplasmosis had significantly higher blood glucose levels during the oral glucose tolerance test (n = 191, p = 0.010; the level of fasting plasma glucose: mean = 5.04 mmol/l vs mean = 4.88 mmol/l; blood glucose level at 1 hour mean = 7.73 mmol/l vs mean = 6.89 mmol/l and blood glucose level at two hours mean = 6.43 mmol/l vs mean = 5.74 mmol/l) and higher prevalence (19.5 %) of gestational diabetes mellitus (n = 532, p = 0.033, odds ratio = 1.78) in the 24–28th gestational weeks than T. gondii- free women (12.0 %). Increased level of glucose and increased incidence of gestational diabetes mellitus could have considerable clinical impact as contributors to the development of the metabolic syndrome and type 2 diabetes in T. gondii -infected women. Our results also brought the first empirical support for the hypothesis that the glucose concentration may play a role in T. gondii -associated offspring sex ratio shifts

The influence of latent toxoplasmosis on women’s reproductive function: four cross-sectional studies

Several studies have investigated the association between infection with Toxoplasma gondii (Nicolle et Manceaux, 1908), pregnancy and fertility, but the results of studies focused on the fertility are rather ambiguous. Here we report results of four new cross-sectional studies. The studies were performed in the General University Hospital, Prague (study A with n = 1 165, and study C with n = 317), in private clinics of the Centre of Reproductive Medicine, Prague (study B with n = 1 016), and in a population of Czech and Slovak volunteers from the Facebook page 'Guinea Pigs' willing to participate in various basic science studies (study D with n = 524). In studies A and B, the clinical records were used to assess the fertility problems, whereas in studies C and D, the women were asked to rate their fertility problems using a six-point scale. Pregnant T. gondii-infected women were older than T. gondii-free women (study A: 33.1 vs 31.2, P < 0.001; study B: 30.6 vs 29.6, P = 0.012) and more often used assisted reproductive technology to conceive (study A: 17.2% vs 12.4%, P = 0.041; study B: 13.4% vs 9.2%, P = 0.317). Pregnant T. gondii-infected primiparous women were older than T. gondii-free primiparas (study A: 31.1 vs 29.5, P < 0.001; study B: 29.7 vs 28.9, P = 0.064) and more often used assisted reproductive technology to conceive (study A: 24.7% vs 14.4%, P = 0.010; study B: 15.9% vs 15.5%, P = 0.888). T. gondii-infected women reported to take a longer time to conceive than T. gondii-free women (P = 0.015). They also claimed to have more fertility problems than T. gondii-free women (P < 0.0001). Our results suggest that 'asymptomatic' latent toxoplasmosis could be a more serious source of fertility problems and health-associated burden than more severe but far rarer congenital toxoplasmosis.

The parasite Toxoplasma gondii has been found to manipulate the behavior of its secondary hosts to increase its own dissemination which is commonly believed to be to the detriment of the host (manipulation hypothesis). The manipulation correlates with an up-regulation of dopaminergic neurotransmission. In humans, different pathologies have been associated with T. gondii infections but most latently infected humans do not seem to display overt impairments. Since a dopamine plus does not necessarily bear exclusively negative consequences in humans, we investigated potential positive consequences of latent toxoplasmosis (and the presumed boosting of dopaminergic neurotransmission) on human cognition and behavior. For this purpose, we focused on action cascading which has been shown to be modulated by dopamine. Based on behavioral and neurophysiological (EEG) data obtained by means of a stop-change paradigm, we were able to demonstrate that healthy young humans can actually benefit from latent T. gondii infection as regards their performance in this task (as indicated by faster response times and a smaller P3 component). The data shows that a latent infection which is assumed to affect the dopaminergic system can lead to paradoxical improvements of cognitive control processes in humans.

Increased rates of exposure to Toxoplasma gondii have been found in individuals with schizophrenia and bipolar disorder, but the association between Toxoplasma and cognitive functioning has not previously been examined. We measured IgG and IgM class antibodies to Toxoplasma in 408 nonelderly individuals with schizophrenia, 347 with bipolar disorder, and 352 nonpsychiatric controls. Cognitive functioning was measured with the Repeatable Battery for the Assessment of Neuropsychological Status. Multivariate linear and regression analyses showed significant associations between Toxoplasma IgM antibody level and cognitive scores within the control group and the bipolar disorder group but not the schizophrenia group. Within the control group, having an elevated Toxoplasma IgM antibody level, greater than or equal to the 50th and 75th levels of the control group, was associated with significantly elevated odds of a low total cognitive score. Exposure to Toxoplasma may confer risk for lower cognitive functioning in persons without a psychiatric disorder and those with bipolar disorder.

BACKGROUND: Schizophrenia is a major psychiatric disorder with a deeply destructive pathophysiology. There are evidences to indicate that infectious agents such as Toxoplasma gondii may play some roles in etiology of the disorder. OBJECTIVES: The current study aimed to determine the association between T. gondii exposure and the risk of schizophrenia. MATERIALS AND METHODS: T. gondii IgG antibodies of 100 patients with schizophrenia as well as 200 healthy volunteers were assessed. The subjects also completed demographic questionnaires. Data was analyzed using the chi-square and Fisher exact tests. RESULTS: The analyses confirmed the significant differences between healthy women and ones with schizophrenia (P = 0.001) as well as between males and females with schizophrenia (P = 0.009) in IgG positivity. CONCLUSIONS: The present study supported the contamination with T. gondii as a risk factor for schizophrenia just in women.

Does prevalence of latent toxoplasmosis correlate with nation-wide rate of traffic accidents?

Latent toxoplasmosis is probably the most common protistan parasitic disease with many indirect negative impacts on human health. One of the important impacts is impaired psychomotor function leading to reduced driving efficiency in Toxoplasma - seropositive subjects. Numerous case-control studies have established a positive relation between the seroprevalence of Toxoplasma gondii (Nicolle et Manceaux, 1908) and probability of traffic accidents in study populations. The prevalence of toxoplasmosis varies between populations according to local geographical conditions, hygienic practices and kitchen habits. Similarly, we see a striking variation in the incidence of traffic accidents across countries. Hence, we compiled the largest ever data set on the seroprevalence of toxoplasmosis and tried to understand its role in traffic accident-related deaths and disabilities across 87 countries. Simple non - parametric analysis showed a positive and strong relation of T. gondii seroprevalence and traffic accident related disabilities. Further, we conducted multivariate analysis to control for confounding factors. After controlling for wealth, geographical latitude, health of population, length of roads and number of vehicles, the correlation disappeared. When the frequency of RhD negativity and its in - teraction with toxoplasmosis were included into the model, the effects of toxoplasmosis seemingly returned. However, the analysed data suffered from the problem of multicollinearity. When a proper method of analysis, ridge regression, was applied, the effects of toxoplasmosis prevalence and RhD negativity frequency disappeared again. The existence of a strong correlation between the prevalence of toxoplasmosis and health of population in particular countries, which was the probable cause of multicollinearity and possible reason for the negative result of the present study, suggests that ‘asymptomatic’ latent toxoplasmosis could have a large impact on public health

Effect of toxoplasmosis on personality profiles of Iranian men and women

There is evidence to suggest that the protozoan Toxoplasma gondii, which causes toxoplasmosis, changes the personality of people who are infected with it. The aim of this study was to compare the personality characteristics of Iranian students with and without latent toxoplasmosis. A total of 237 students (111 men and 126 women) of Jundishapur University of Medical Sciences (Ahvaz, Iran) were tested for the presence of anti-Toxoplasma antibodies and completed demographic questionnaires and Cattell's 16 Personality Factor Questionnaire. Data were analysed using multiple univariate analyses of variance. Women with latent toxoplasmosis had a significantly different personality profile from women without toxoplasmosis, namely higher O (apprehension), N (privateness) and Q(4) (tension) scores, and lower Q(1) (openness to change) scores. Infected men had significantly higher L (vigilance, mistrust) scores compared to non-infected men. Factors E (dominance) and Q(1) (openness to change) tended to be higher in infected men than non-infected men but the difference was not quite statistically significant. Our findings have, for the first time, independently confirmed that personality profile is affected by latent toxoplasmosis.

Influence of latent Toxoplasma infection on human personality, physiology and morphology: pros and cons of the Toxoplasma-human model in studying the manipulation hypothesis

The parasitic protozoan Toxoplasma gondii infects about one-third of the population of developed countries. The life-long presence of dormant stages of this parasite in the brain and muscular tissues of infected humans is usually considered asymptomatic from the clinical point of view. In the past 20  years, research performed mostly on military personnel, university students, pregnant women and blood donors has shown that this ʻ asymptomatic ʼ disease has a large influence on various aspects of human life. Toxoplasma -infected subjects differ from uninfected controls in the personality profile estimated with two versions of Cattell ʼ s 16PF, Cloninger ʼ s TCI and Big Five questionnaires. Most of these differences increase with the length of time since the onset of infection, suggesting that Toxoplasma influences human personality rather than human personality influencing the probability of infection. Toxoplasmosis increases the reaction time of infected subjects, which can explain the increased probability of traffic accidents in infected subjects reported in three retrospective and one very large prospective case-contr ol study. Latent toxoplasmosis is associated with immunosuppression, which might explain the increased probability of giving birth to a boy in Toxoplasma- infected women and also the extremely high prevalence of toxoplasmosis in mothers of children with Down syndrome. Toxoplasma -infected male students are about 3  cm taller than Toxoplasma- free subjects and their faces are rated by women as more masculine and dominant. These differences may be caused by an increased concentration of testosterone. Toxoplasma also appears to be involved in the initiation of more severe forms of schizophrenia. At least 40 studies confirmed an increased prevalence of toxoplasmosis among schizophrenic patients. Toxoplasma- infected schizophrenic patients differ from Toxoplasma- free schizophrenic patients by brain anatomy and by a higher intensity of the positive symptoms of the disease. Finally, five independent studies performed in blood donors, pregnant women and military personnel showed that RhD blood group positivity, especially in RhD heterozygotes, protects infected subjects against various effects of latent toxoplasmosis, such as the prolongation of reaction times, an increased risk of traffic accidents and excessive pregnancy weigh t gain. The modern human is not a natural host of Toxoplasma . Therefore, it can only be speculated which of the observed effects of latent toxoplasmosis are the result of the manipulation activity of the Toxoplasma aimed to increase the probability of its transmission from a natural intermediate to the definitive host by predation, and which are just side effects of chronic infection

Parasite location has been proposed as an important factor in the behavioural changes observed in rodents infected with the protozoan Toxoplasma gondii. During the chronic stages of infection, encysted parasites are found in the brain but it remains unclear whether the parasite has tropism for specific brain regions. Parasite tissue cysts are found in all brain areas with some, but not all, prior studies reporting higher numbers located in the amygdala and frontal cortex. A stochastic process of parasite location does not, however, seem to explain the distinct and often subtle changes observed in rodent behaviour. One factor that could contribute to the specific changes is increased dopamine production by T. gondii. Recently, it was found that cells encysted with parasites in the brains of experimentally infected rodents have high levels of dopamine and that the parasite encodes a tyrosine hydroxylase, the rate-limiting enzyme in the synthesis of this neurotransmitter. A mechanism is proposed that could explain the behaviour changes due to parasite regulation of dopamine. This could have important implications for T. gondii infections in humans

oxoplasmosis, a zoonosis caused by a protozoan, Toxoplasma gondii , is probably the most widespread human parasitosis in developed countries. Pregnant women with latent toxoplasmosis have seemingly younger fetuses especially in the 16thweekof gestation,which suggests that fetuses of Toxoplasma -infected mothers have slower rates of development in the fi rst trimester of pregnancy. In the present retrospective cohort study, we analyzed data on postnatal motor development of infants from 331 questionnaire respondents including 53 Toxoplasma - infected mothers to search for signs of early postnatal development disorders. During the fi rst year of life, a slower postnatal motor development was observed in infants of mothers with latent toxoplasmosis. These in- fants signi fi cantly later developed the ability to control the head position (p=0.039), to roll from supine to prone position (p=0.022) and were slightly later to begin crawling (p=0.059). Our results are compatible with the hypothesis that the difference in the rates of prenatal and early postnatal development between chil- drenof Toxoplasma -negative and Toxoplasma -positive mothers might becausedbya decreased stringency of em- bryo quality control in partly immunosuppressed Toxoplasma -positive mothers resulting in a higher proportion of infants with genetic or developmental disorders in offspring. However, because of relatively low return rate of questionnaires and an associated risk of a sieve effect, our results should be considered as preliminary and performing a large scale prospective study in the future is critically needed.

oxoplasmosis is associated with speci fi c differences in the personality of infected subjects relative to non-infected subjects. These differences are usually cons idered to be a side effect of the manipulative activity of the parasite aimed to increase the probability of its transmission from the intermediate host to the de fi nitive host by predation. The personality of infected s ubjects was studied mostly using the Cattell ’ s questionnaire. However, this questionnaire is now considered outdated and has been mostly substituted with the Neuroticism – Extraversion – Openness Personality Inventory — Revised (NEO-PI-R) questionnaire in clinical practice. Here, we searched for the as- sociation between toxoplasmosis and the personality by screening a population of students with the NEO-PI-R question- naire. We found that Toxoplasma-infected male and female students had signi fi cantly higher extraversion and lower conscientiousness. The conscientiousness negatively correlated with the length of infection in men, which suggested that the toxoplasmosis associated differences were more probably the result of slow cumulative changes induced by latent toxoplasmosis, rather than transient side effect of acute Toxoplasma infection. The existence of this correlation also sup- ported (but of course not proved) the hypothesis that Toxoplasma infection in fl uenced the personality, rather than the hypothesis that the personality in fl uenced the probability of the infection.

Worldwide, approximately 2 billion people are chronically infected with Toxoplasma gondii with largely yet unknown consequences. Patients with autism spectrum disorders (ASD) similarly as mice with chronic toxoplasmosis have persistent neuroinflammation, hypercytokinemia with hypermetabolism associated with enhanced lipid peroxidation, and extreme changes in the weight resulting in obesity or wasting. Data presented in this review suggest that environmental triggering factors such as pregnancy, viral/bacterial infections, vaccinations, medications, and other substances caused reactivation of latent cerebral toxoplasmosis because of changes in intensity of latent central nervous system T. gondii infection/inflammation and finally resulted in development of ASD. Examples of such environmental factors together with their respective biomarker abnormalities are: pregnancy (increased NO, IL-1 beta, TNF-alpha, IL-6, IL-10, prolactin: decreased IFN-gamma, IL-12), neuroborreliosis (increased IL-1 beta, sIL-1R2, TNF-alpha, IFN-gamma, IL-6, IL-10, IL-12, IL-18, transforming growth factor-beta 1 (TGF-beta 1)), vital infections (increased IL-1 beta, IL-6, IL-8, TNF-alpha, IFN-gamma/alpha/beta,TGF-beta 1), thimerosal (increased IL-5, IL-13; decreased IFN-gamma,TNF-alpha,IL-6, IL-12p70, NOS), and valproic acid (increased NO, reactive oxygen species; decreased TNF-alpha, IL-6, IFN-gamma). The imbalances in pro- and antiinflammatory processes could markedly hinder [lost defense mechanisms important for immune control of the parasite, such as the production of NO, cytokines, and reactive oxygen/nitrogen species, tryptophan degradation by indoleamine 2,3-dioxygenase and/or tryptophan 2,3-dioxygenase, limitation of the availability of intracellular iron to T gondii, and the mechanisms mediated by an IFN-gamma responsive gene family. These fluctuations could result in a recurrent profuse multiplication of T. gondii in the brain associated with persistent neuroinflammation, chronic overproduction of pro- and antiinflammatory cytokines, and NO causing increased oxidative stress, and significantly depressed activity of several enzymes including cytochrome P450 monooxygenase family responsible for metabolism of physiological substrates and xenobiotics, such as steroids, fatty acids, prostaglandins, drugs, pollutants, and carcinogens, finally leading to development of ASD. This reasoning may be supported by such abnormal metabolic events as: (1) patients with ASD have significantly decreased melatonin levels caused by marked deficit in acetylserotonin methyltransferase activity, possibly resulting from maternal and/or fetal/postnatal overproduction of NO, characteristic for this clinical entity; (2) thimerosal inhibited both insulin-like growth factor-1- and dopamine-stimulated methylation reactions, and depressed methionine synthase activity, the metabolic events important for promoting normal neurodevelopment; (3) valproic acid, a strong histone deacetylase inhibitor, have potent anti-T. gondii activity. The influence of RhD phenotype on toxoplasmosis- and age-associated changes in personality profile of blood donors

Toxoplasma gondii , a parasite infecting 20–60% of humans in various countries, influences the behaviour of infected animal and human hosts. Infected human subjects have changes in several of Cattell’s and Cloninger’s personality factors. Recently, three independent studies have shown that Rh-positive subjects are protected against the T. gondii -induced changes of reaction times and increased risk of traffic accidents. Here we searched for evidence of similar effects of RhD phenotype on toxoplasmosis- or aging-associated changes in the personality profile of about 302 blood donors. We found that Rh-positive and Rh-negative subjects responded differently to toxoplasmosis. In addition to the already known effects of toxoplasmosis on novelty seeking, self transcend - ence, superego strength and protension, we also found effects of RhD phenotype on ego strength, protension, and praxernia, as well as opposite effects of toxoplasmosis on ego strength, praxernia, ergic tension and cooperativeness in Rh-positive and Rh-negative subjects. Moreover, our results indicate that RhD phenotype might influence not only the effect of toxoplasmosis but also the effect of aging on specific personality traits.

The RhD protein which is the RHD gene product and a major component of the Rh blood group system carries the strongest blood group immunogen, the D-antigen. This antigen is absent in a significant minority of the human population (RhD-negatives) due to RHD deletion or alternation. The origin and persistence of this RhD polymorphism is an old evolutionary enigma. Before the advent of modern medicine, the carriers of the rarer allele (e.g. RhD-negative women in the population of RhD-positives or RhD-positive men in the population of RhD-negatives) were at a disadvantage as some of their children (RhD-positive children born to pre-immunized RhD-negative mothers) were at a higher risk of foetal or newborn death or health impairment from haemolytic disease. Therefore, the RhD-polymorphism should be unstable, unless the disadvantage of carriers of the locally less abundant allele is counterbalanced by, for example, higher viability of the heterozygotes. Here we demonstrated for the first time that among Toxoplasma -free subjects the RhD-negative men had faster reaction times than Rh-positive subjects and showed that heterozygous men with both the RhD plus and RhD minus alleles were protected against prolongation of reaction times caused by infection with the common protozoan parasite Toxoplasma gondii . Our results suggest that the balancing selection favouring heterozygotes could explain the origin and stability of the RhD polymorphism. Moreover, an unequal prevalence of toxoplasmosis in different countries could explain pronounced differences in frequencies of RhD-negative phenotype in geographically distinct populations

BACKGROUND: The biological function of RhD protein, a major component of the Rh blood group system, is largely unknown. No phenotypic effect of RhD protein, except its role in hemolytic disease of newborns and protective role against Toxoplasma-induced impairment of reaction times in men, has been described. METHODS: Here we searched for a protective effect of RhD positivity against Toxoplasma-induced prolongation of reaction times in a set of 110 male and 226 female students of the Faculty of Science tested for latent toxoplasmosis and concentration of testosterone in saliva. RESULTS: RhD-positive subjects have been confirmed to be less sensitive to the influence of latent toxoplasmosis on reaction times than Rh-negative subjects. While a protective role of RhD positivity has been demonstrated previously in four populations of men, the present study has shown a similar effect in 226 female students. Our results have also shown that the concentration of testosterone in saliva strongly influences (reduces) reaction times (especially in men) and therefore, this factor should be controlled in future reaction times studies. CONCLUSIONS: The observed effects of RhD phenotype could provide not only a clue to the long-standing evolutionary enigma of the origin of RhD polymorphism in humans (the effect of balancing selection), differences in the RhD+ allele frequencies in geographically distinct populations (resulting from geographic variation in the prevalence of Toxoplasma gondii), but might also be the missing piece in the puzzle of the physiological function of the RhD molecule.

Background: Increased rates of exposure to Toxoplasma gondii have been found in individuals with schizophrenia as compared with control groups, but the correlates of Toxoplasma exposure in schizophrenia have not been defined. Methods: We measured IgG class antibodies to Toxoplasma gondii in 358 individuals with schizophrenia. We correlated Toxoplasma antibody status with clinical and demographic variables and examined the effect of Toxoplasma seropositivity on mortality in a follow-up period of up to 5 years. Results: Individuals with schizophrenia who had serological evidence of Toxoplasma infection were more likely to be female but did not differ in age, race, total symptom score, or other demographic or clinical characteristics. However, we found that serological evidence of Toxoplasma was associated with a significantly increased risk of dying of natural causes during the follow-up period (Cox proportional hazard ratio of 4.70; 95% confidence interval, 1.27-17.31, P =.020) adjusted for age, gender, and other clinical and demographic variables. Conclusions: Toxoplasma infection may confer an increased risk for mortality from natural causes in schizophrenia. An understanding of the pathogenesis of Toxoplasma infections in individuals with schizophrenia might lead to new approaches to the management of this disorder.

Toxoplasma is parasite of cats that uses any warm-blooded animals as intermediate hosts. It is known to induce shifts in behavior, physiology and even morphology of its intermediate hosts, including humans. The lower second to fourth digit ratio (2D:4D ratio) in infected man and women, and higher height in infected man sug - gest that sex steroid hormones like testosterone could play a role in these shifts. Here, we searched for another indirect indication for a higher postnatal testosterone level, i.e. increased perceived dominance and masculinity in infected men. We showed portrait pictures of 89 male students of which 18 were Toxoplasma - infected to 109 female students. When we statistically corrected for age, men with latent toxoplasmosis were perceived as more dominant (p=0.009) and masculine (p=0.052). These results support the idea that the higher level of testosterone could be responsible for at least some of the toxoplasmosis-associated shifts in human and animal behavior.

OBJECTIVES : Toxoplasma gondii , a parasitic protozoan, infects about 30–60% of people worldwide. Toxoplasma is known to induce behavioral changes and an increase of dopamine in mice. The presence of anti- Toxoplasma antibodies (latent toxoplasmosis) is also a risk factor for schizophrenia. Latent toxoplasmosis in men (male soldiers) is associated with lower novelty seeking. As the novelty seek - ing is supposed to negatively correlate with level of dopamine, the observed effect was interpreted as indirect evidence of increased dopamine levels in subjects with toxoplasmosis. However, it is also possible that the observed effect was caused by association of both novelty seeking and Toxoplasma infection with a third factor, e.g. size of place of residence. METHODS : Personality profile of 290 blood donors (205 men and 85 women) were measured by Cloninger’s TCI (Temperament and Character Inventory) and their blood samples were assayed for the presence of anti- Toxoplasma antibodies. Dif - ference between Toxoplasma -infected and Toxoplasma -free subjects was tested with ANCOVA method with gender, size of place of residence, and age as covari - ates. RESULTS : The present analysis revealed that lower novelty seeking was associ - ated with latent toxoplasmosis both in men and women. The effect of infection on novelty seeking remained significant even after adjustment for size of place of residence (p<0.0). CONCLUSION : Decreased novelty seeking in Toxoplasma -infected subjects have been already confirmed in three independent populations (male soldiers and male and female blood donors). These findings suggest that the local inflamma - tion-induced increase in dopamine in the brain of infected subjects can represent a missing link between toxoplasmosis and schizophrenia

Here we searched for symptoms of de terioration of physical health (decrease of weight) in infected people by analysis o f clinical records of 758 women tested for toxoplasmosis in the 16th week of gravidity. Toxoplasma -positive women have a lower body weight in the 16th week of gravidity (p = 0.02) than Toxoplasma -negative women. Moreover, a negative correlation be- tween weight and the duration of toxoplasmosis was found in a subset of 174 Toxoplasma -positive women (p = 0.04), suggesting that slow and cumulative effects of latent toxoplasmosis, rather than a transient eff ect of acute toxoplasmosis, are responsibl e for the decreased weight of infected subjects. Longer duration of gr avidity estimated from the date of last menstruation in the set of Toxoplasma -positive women in the 16th week of gravidity estimated with ultrasonography (p = 0.04) suggests a possibility of retarded foetal growth in Toxoplasma -positive women. The prevalence of latent toxopl asmosis is extremely high. Therefore, even its mild symptoms such as the decreased body weight in Toxoplasma- positive pregnant women mi ght in fact indicate an unrecognized serious public health problem.

The exact mechanisms of action of some antipsychotics and mood stabilizers have not been elucidated. Response to these medications can vary among individuals. Recent studies indicate that infection with the parasite Toxoplasma gondii may contribute to the symptoms of schizophrenia in some individuals. We investigated commonly used antipsychotic and mood stabilizing medications for their ability to inhibit the replication of this organism. We employed a system for testing compounds for in vitro activity against T gondii. Human fibroblasts (HFF) were treated with test compounds and then exposed to Toxoplasma that has been genetically modified to express cytoplasmic galactosidase. Inhibition by the drugs was determined by spectrophotometric analysis of colorimetric reactions. We tested 12 neuroleptic compounds and found that of these, the antipsychotic haloperidol and the mood stabilizer valproic acid most effectively inhibit Toxoplasma growth in vitro. Valproic acid inhibited the parasite at a concentration below that found in the cerebrospinal fluid and blood of individuals being treated with this medication and displayed synergistic activity with haloperidol and with trimethoprim, an antibiotic commonly used to treat Toxoplasma infections. Several medications used to treat schizophrenia and bipolar disorder have the ability to inhibit the in vitro replication of T gondii. (C) 2002 Elsevier Science B.V. All rights reserved.

Toxoplasma gondii , a parasitic protozoan, infects about 30  / 60% of people worldwide. The latent toxoplasmosis, i.e. life-long presence of cysts in the brain and muscular tissues, has no effect on human health. Howe v er, infected subjects score worse in psychomotor performance tests and ha v e different personality profiles than Toxoplasma -negati v e subjects. The mechanism of this effect is unknown; howe v er, it is supposed that presence of parasites’ cysts in the brain induces an increase of the concentration of dopamine. Here we search for the existence of differences in personality profile between Toxoplasma -positi v e and Toxoplasma - negati v e subjects by testing 857 military conscripts using a modern psychobiological questionnaire, namely with Cloninger’s Temperament and Character In v entory (TCI). ANCOVA showed that Toxoplasma -positi v e subjects had lower No v elty seeking (NS) scores( P  / 0.035) and lower scores for three of its four subscales, namely Impulsi v eness ( P  / 0.049), Extra v agance ( P  / 0.056) and Disorderliness ( P  / 0.006) than the Toxoplasma -negati v e subjects. Differences between Toxoplasma -negati v e and positi v e subjects in NS was in v ersely correlated with duration of toxoplasmosis estimated on the basis of concentration anti- Toxoplasma antibodies ( P  / 0.031). Unexpectedly, the infected subjects had also lower IQ ( P 2  / 0.003) and lower probability of achie v ing a higher education ( P 2 B / 0.0000). Decrease of NS suggests that the increase of dopamine in brain of infected subjects can represent a missing link between toxoplasmosis and schizophrenia

The manipulation hypothesis states a parasite may alter host behaviour for its own benefit, often by enhancing its transmission rate through the food chain. This paper reviews studies on the potential impact of one parasite, Toxoplasma gondii, on host behaviour, both on rodents, where altered responses may be proposed to benefit the parasite, and humans, where altered responses may arise as a side-effect of infection with no current adaptive significance. Toxoplasma gondii is known to induce speciÆc behavioural changes in its intermediate hosts. This is usually considered to be an evolutionary adaptation aimed to increase the probability of transmission of the parasite into its deÆnitive host, the cat, by predation. In rodents an increase of reaction time as well as many other speciÆc behavioural patterns have been observed. Here we report the results of our double blind study showing the signiÆcantly longer reaction times of 60 subjects with latent toxoplasmosis in comparison with those of 56 controls. Moreover, the existence of a positive correlation between length of infection and mean reaction time suggested that slow and cumulative effects of latent toxoplasmosis rather than a one-step (and possibly transient) effect of acute toxoplasmosis disease are responsible for the decrease of psychomotor performance of infected subjects. To our knowledge, this is the Ærst study conÆrming the existence of such parasite-induced changes in human behaviour that could be considered in evolutionary history of the human species as adaptive from the point of view of parasite transmission.

Correlation of duration of latent Toxoplasma gondii infection with personality changes in women

Many parasites induce characteristic changes in behavior of their hosts. In humans latent toxoplasmosis is associated with changes in personality profiles. It has been already shown that a decrease in superego strength is correlated with duration of toxoplasmosis in men. Here we studied changes in personality profiles with Cattell’s 16 PF questionnaire in Toxoplasma -infected women. The changes were measured as differences in personality factors between Toxoplasma -infected subjects and uninfected controls of the same age. The low-rate changes were studied in 230 women diagnosed with acute toxoplasmosis during past 14 years. The results showed the correlation between duration of toxoplasmosis and level of factors G (high superego strength) and Q3 (high strength of self sentiment). The high-rate changes were estimated by measuring the correlation between level of Toxoplasma -antibody titers (which rapidly decline after the end of acute phase of toxoplasmosis) and personality factors in an experimental set of 55 young mothers with latent toxoplasmosis. Again, certain factors, namely A (affectothymia), F (surgence), G (high superego strength), H (parmia), and L (protension), correlated with the length of the infection. We suggest that the parasite induced the changes in the personality profiles of the women because of our observation of an increasingly different personality profile over time between women with latent infection and controls. The same evidence questions the view that women with a particular personality profile are more prone to acquisition
