Toxoplasma gondii IgG associations with sleep-wake problems, sleep duration and timing

Background: Evidence links Toxoplasma gondii (T. gondii), a neurotropic parasite, with schizophrenia, mood disorders and suicidal behavior, all of which are associated and exacerbated by disrupted sleep. Moreover, low-grade immune activation and dopaminergic overstimulation, which are consequences of T. gondii infection, could alter sleep patterns and duration. Methods: Sleep data on 833 Amish participants [mean age (SD) = 44.28 (16.99) years; 59.06% women] were obtained via self-reported questionnaires that assessed sleep problems, duration and timing. T. gondii IgG was measured with ELISA. Data were analyzed using multivariable logistic regressions and linear mixed models, with adjustment for age, sex and family structure. Results: T gondii seropositives reported less sleep problems (p < 0.005) and less daytime problems due to poor sleep (p < 0.005). Higher T. gondii titers were associated with longer sleep duration (p < 0.05), earlier bedtime (p < 0.005) and earlier mid-sleep time (p < 0.05). Conclusions: It seems unlikely that sleep mediates the previously reported associations between gondii and mental illness. Future longitudinal studies with objective measures are necessary to replicate our findings.

Humans infected with the parasite Toxoplasma gondii display a wide variety of abnormal behaviours, from suicide and depression to stuttering. These behaviours have been seen as so serious as to constitute a public health problem. It is not clear to what extent the parasite is a cause of, or merely a marker for, these behaviours, but there is evidence for both. Some of these behaviours are associated with changes in steroid hormones, that is, estrogen in women and testosterone in men. It is suggested here that these endocrine-related states of infected people may be better understood by studying their offspring sex ratios.

The association of latent toxoplasmosis with mental disorders in general and with schizophrenia in particular was noticed in the mid-1950s. In subsequent years, the role of Toxoplasma gondii was established based on its ability to survive for long periods of time in the nerve cells of the brain. The acute manifestations of the infection include psychopathic symptoms resembling those of schizophrenia. In the former USSR, and in other parts of the world, a number of studies were performed with respect to the association of latent toxoplasmosis and schizophrenia. However, with the dissolution of the USSR at the beginning of the 1990s, studies on the subject were halted due to financial problems and have resumed only recently. The reasons for the resumption of such studies in contemporary Russia are related to the progressively increasing incidence of schizophrenia over the last 25-30 years in the country. According to official data, approximately 550 000 persons reported suffering from the disease in 2014. There are reasons to believe that this is only a fraction of the real burden of the disease. Economically, it cost the state no less than approximately US $10 billion. The purpose of the study was to identify the level of toxoplasmosis seroprevalence in patients with verified diagnoses of schizophrenia in comparison to healthy people in Moscow City and in the Moscow region. A total of 155 persons constituted the patients group and 152 healthy people were in the control group. An integrated approach to the diagnosis and comparison of data from the entire spectrum of serological markers of infection was used, including the detection of specific IgM and the determination of IgG concentrations. It was found that among persons with neuropsychiatric disorders, the incidence of cases with latent toxoplasmosis was higher than in the control group. The effect of toxoplasmosis was significant and similar for men and women. Further statistical analyses revealed that among patients with a diagnosis of schizophrenia, the incidence of latent toxoplasmosis was significantly higher than in the control group. These data are in agreement with the results of similar studies in other countries.

Economic holobiont: Influence of parasites, microbiota and chemosignals on economic behavior

The article is a perspective on utilization of microorganisms and chemosignals in studying human economic behavior. Research in biological roots of economic development has already confirmed that parasitic pressure influenced the creation and development of cultural norms and institutions. However, other effects of microorganisms on human groups and individual decision-making and behavior are heavily understudied. The perspective discusses how parasitic infections, sexually transmitted organisms and microbiota (i.e., "human holobiont") could causally influence risk-seeking behavior, impulsivity, social dominance, empathy, political views and gender differences. As a case study, the parasite Toxoplasma gondii and its influence on economic preferences, personal characteristics and human appearance are examined. I also briefly review how chemosignals influence decision-making, particularly in the social preferences domain. I mention some predictions that arise from the paradigm of economic holobiont for the economic science. The conclusion summarizes limitations of the discussed findings and the stated speculations.

Depressiveness and neuroticism in Bartonella seropositive and seronegative subjects-preregistered case-controls study

Several recent studies have demonstrated the association of cat-related injuries with major depression and with depressiveness in the general population. It was suggested that cat-scratch disease, the infection with the bacterium Bartonella henselae, can be responsible for the observed association. However, no direct evidence for the role of the Bartonella infection in this association has been published until now. In this preregistered case-controls study performed on 250 healthy subjects tested earlier for the presence of anti-Toxoplasma IgG antibodies, we searched for the positive association between presence of anamnestic anti-Bartonella IgG antibodies and depressiveness measured with Beck II inventory, depression subscale of neuroticism measured with N-70 questionnaire, and self-reported health problems. We found that that Bartonella seropositivity was positively correlated with Beck depression only in Toxoplasma-seronegative men and negatively correlated with health in Toxoplasma-seronegative women. Bartonella seropositivity expressed protective effects against Toxoplasma seropositivity-associated increased neuroticism in men while Toxoplasma-seropositivity expressed protective effects against Bartonella seropositivity-associated health problems in women. A comparison of the patterns of association of mental and physical health problems with Bartonella seropositivity and with reported cat-related injury suggests that different factor, possibly infection with different pathogen transmitted by cat related-injuries than the B. henselae, is responsible for the observed association of cat related-injuries with depressiveness and major depression. The existence of complex interactions between Bartonella seropositivity, Toxoplasma seropositivity, and sex also suggest that the effect of symbionts on the host's phenotype must by always studied in the context of other infections, and separately for men and women.

Lower performance of Toxoplasma-infected, Rh-negative subjects in the weight holding and hand-grip tests

Background Toxoplasma, a protozoan parasite of cats, infects many species of intermediate and paratenic hosts, including about one-third of humans worldwide. After a short phase of acute infection, the tissue cysts containing slowly dividing bradyzoites are formed in various organs and toxoplasmosis proceeds spontaneously in its latent form. In immunocompetent subjects, latent toxoplasmosis was considered asymptomatic. However, dozens of studies performed on animals and humans in the past twenty years have shown that it is accompanied by a broad spectrum of specific behavioural, physiological and even morphological changes. In human hosts, the changes often go in the opposite direction in men and women, and are mostly weaker or non-existent in Rh-positive subjects. Methods Here, we searched for the indices of lower endurance of the infected subjects by examining the performance of nearly five hundred university students tested for toxoplasmosis and Rh phenotype in two tests, a weight holding test and a grip test. Results The results confirmed the existence of a negative association of latent toxoplasmosis with the performance of students, especially Rh-negative men, in these tests. Surprisingly, but in an accordance with some already published data, Toxoplasma-infected, Rh-positive subjects expressed a higher, rather than lower, performance in our endurance tests. Discussion Therefore, the results only partly support the hypothesis for the lower endurance of Toxoplasma infected subjects as the performance of Rh-positive subjects (representing majority of population) correlated positively with the Toxoplasma infection.

The relation of secondary sex ratio and miscarriage history with Toxoplasma gondii infection

Background: Toxoplasma gondii is a protozoan parasite with worldwide distribution, infecting a broad-range of humans and warm-blooded animals. In the current study, role of this parasite on secondary sex ratio and risk of miscarriage was investigated. Methods: In this cross-sectional study, 850 cord blood samples were collected in Tehran, Iran, 2014-2015. Enzyme-linked immunosorbent assay (ELISA) was used to assess anti-Toxoplasma IgG in samples. Information such as sex of the neonates and age, number of previous pregnancies and history of miscarriage of the mothers were recorded in questionnaires. Logistic regression analysis was used to assess the possible relationship between the latent toxoplasmosis and the highlighted parameters. Results: Logistic regression analysis showed that the odds of having a male neonate in seropositive women is nearly 64% higher than that in seronegative women (OR = 1.64, CI95 = 1.16-2.33, P = 0.005). The odds ratio of having male neonate increased to 2.10 (CI95 = 1.24-3.57, P = 0.006) in high-titer seropositive women, compared to that in seronegative control group. The odds of having a miscarriage history was approximately two and a half times greater in seropositive women than in seronegative ones (OR = 2.45, CI95 = 1.56-3.87, P < 0.001). The odds ratio of having miscarriage increased to 2.76 (CI95 = 1.61-4.73, P < <.001) in low-titer seropositive women, compared to that in seronegative control group. Conclusion: Results of the current study have shown that T. gondii infection affects secondary sex ratio in human offspring and can be addressed as one of the major miscarriage causes in women.

Differences in cognitive functions between cytomegalovirus-infected and cytomegalovirus-free university students: a case control study

Cytomegalovirus (CMV) is the herpetic virus, which infects 45-100% people worldwide. Many reports suggest that CMV could impair cognitive functions of infected subjects. Here we searched for indices of effects of CMV on infected subjects' intelligence and knowledge. The Intelligence Structure Test I-S-T 2000 R was used to compare IQ of 148 CMV-infected and 135 CMV-free university students. Infected students expressed higher intelligence. Paradoxically, their IQ decreased with decreasing concentration of anti-CMV antibodies, which can be used, statistically, as a proxy of the time passed from the moment of infection in young subjects when the age of subjects is statistically controlled. The paradox of seemingly higher intelligence of CMV infected subjects could be explained by the presence of the subpopulation of about 5-10% CMV-positive individuals in the population of "CMV-negative students". These false negative subjects had probably not only the oldest infections and therefore the lowest concentration of anamnestic antibodies, but also the lowest intelligence among the infected students. Prevalence of CMV infection in all countries is very high, approaching sometimes 90%. Therefore, the total impact of CMV on human intelligence may be large.

Studies suggest that the protozoan Toxoplasma gondii can disturb human behavior. This study aimed to systematically review the scientific literature on the possible associations between Toxoplasma gondii infection and neurobehavioral abnormalities in humans. We reviewed and summarized the studies published since 1990. The descriptors used were related to T. gondii infection and behavioral outcomes in humans; the main databases of the medical literature were accessed. The results of eight original articles published between 1994 and 2016 were evaluated and described. The most common serological method was the enzyme immunoassay. Most of the researchers used validated instruments for behavioral evaluation. Seven studies reported some association between the prevalence of anti-T. gondii antibodies and some altered behavioral aspects in adult humans; these studies focused on adult population in Europe and the USA. The most reported behavioral deviations are related to greater impulsivity and aggressiveness. There are very few studies on this subject, which present some limitations for inference and conclusions: most were cross-sectional studies, with a small sample size and in similar populations. Investigations with a larger sample size of different population groups should be performed to evaluate multiple factors.

Background: Evidence suggests that Toxoplasma gondii, which causes toxoplasmosis, changes the metabolism of neurotransmitters, especially dopamine, resulting in both neurologic and psychiatric disorders. On the other hand, the dysregulation of catecholamines, especially of both norepinephrine and dopamine, has been proposed in the pathophysiology of attention deficit hyperactivity disorder (ADHD). The aim of the present study was to investigate anti-toxoplasma antibodies in children and adolescents with ADHD and compare it with a control group, to determine whether toxoplasmosis is a risk factor for ADHD. Methods: A total of 200 children and adolescents (117 patients with ADHD and 83 individuals without ADHD) participated in the study. Participants were tested for the presence of anti-T. gondii antibodies by enzyme-linked immunosorbent assay. A Parent ADHD Rating Scale and Clinical Global Impression-severity Scale were also completed. Data were analyzed using a (2) test and Fisher exact test. Results: Anti-toxoplasma antibodies were detected in 18.1% of patients with ADHD disorder and 24% of individuals without ADHD. There was no significant difference in seropositivity between the 2 groups (P > 0.05). The number of patients with T. gondii infection in the 3 subgroups was 0, 9 and 12, respectively. The differences in infection rate among subgroups were not statistically significant (P > 0.05). Conclusion: Although not conclusive, the present study does not support the theory that T. gondii is a risk factor for ADHD.

Background: The highly prevalent parasitic infection of humans, toxoplasmosis, can have serious consequences even during an asymptomatic course of the disease. The causative agente of infection is Toxoplasma gondii, which proliferates in the intestine of cats. Risk of infection by direct contact with cats is low, but the infection can occur due to contaminated soil, water, unwashed vegetables and fruits, as well as insufficiently heat-treated meat. Objectives: The aim of this study was to synthesize the current knowledge about infection caused by Toxoplasma gondii in a brief review focusing on latent toxoplasmosis in humans. Materials and Methods: More than hundred articles, studies and reports from scientific journals were considered to summarize the infection's characteristics such as prevalence and symptoms in humans. Results and Discussion: The symptoms of acute toxoplasmosis are either featureless, or non-existent, although, for people with immunosuppression it can be fatal, and for expectants it can result in foetal harm. The detection of the infection is conducted with serology methods, however, only in a few European countries are general screenings present. The asymptomatic carrier state is dangerous as well, because the cysts subsist in the brain of the host, which affects behaviour, the regulation of emotions, and also the level of several hormones. The latent toxoplasmosis can be connected to aggression, anxiety, schizophrenia, bipolar personality disorder, suicidal inclination, depression, and the reduction in intelligence quotient. For the reason of the deterioration of reaction time, the disease increases the risk of traffic accidents. The latent toxoplasmosis also influences personality traits; for women and men the alteration of the personality change differs. There is no data on the current epidemiological situation in Hungary, and not even in Europe. A research regarding the extent of asymptomatic carrier state and about the psychological changes in the Hungarian population has not been conducted so far.

OBJECTIVES To investigate the association between schizophrenia and Toxoplasma gondii, and to assess the association of infection with suicide attempts and age of onset of schizophrenia in these patients. METHODS Case-control study Fars Province, southern Iran. Cases were individuals with psychiatric diagnosis of schizophrenia as per Diagnostic and Statistical Manual of Mental Disorders (DSM-IV) criteria. Controls were healthy blood donors, frequency-matched with patients according to age and sex. For the detection of IgG antibodies, enzyme-linked immunosorbent assay (ELISA) was used. Data about demographic information in all subjects and duration of illness and history of suicide attempts in patients with schizophrenia were collected using a brief questionnaire and hospital records. Chi-square test and multivariable logistic regression were used for statistical analyses. RESULTS Among 99 cases, 42 individuals (42%) were positive for T. gondii antibody, vs. 41 (27%) among 152 controls (OR = 2, 95% CI: 1.2-3.4, P = 0.012). We compared the suicide attempts in patients with schizophrenia based on their T. gondii serologic status. There was a lower rate of suicide attempts in seropositive male patients than seronegative ones (OR = 0.3, 95% CI: 0.1-0.97, P = 0.04). Age of onset of schizophrenia did not differ between T. gondii-infected and non-infected patients. CONCLUSIONS These findings may have implications for schizophrenia and suicide prevention programmes. However, clearly further studies are required to confirm them.

Significance of chronic toxoplasmosis in epidemiology of road traffic accidents in Russian Federation

Studies carried out in Moscow residents have revealed that the prevalence of chronic toxoplasmosis is very close to those in countries of Eastern and Central Europe. Our findings also demonstrated a statistically significant relationship between the rate of traffic accidents and the seroprevalence of chronic toxoplasmosis in drivers who were held responsible for accidents. The latter was 2.37 times higher in drivers who were involved in road accidents compared with control groups. These results suggest that the consequences of chronic toxoplasmosis (particularly a slower reaction time and decreased concentration) might contribute to the peculiarities of the epidemiology of road traffic accidents in the Russian Federation and might interfere with the successful implementation of the Federal Programme named "Increase road traffic safety" . Suggestions for how to address overcome this problem are discussed in this paper.

Puppet master: possible influence of the parasite Toxoplasma gondii on managers and employees

The article reviews recent literature on the effects of host manipulation by the parasite Toxoplasma gondii (prevalent in about a third of the world's population) on perception, cognition, and behavior of humans, and on the changes in their physical appearance and personality characteristics. I argue that the mind-affecting parasite paradigm offers many research opportunities for management sciences, especially for organizational psychology and neuroscience. The article summarizes the parasite's physiological mechanisms of affecting the host; highlights important behavioral effects of the infection in humans; and speculates on the possible impacts on skills and careers of employees and managers, organizational dynamics, intercultural management, and gender work roles. The conclusion shows limitations of the presented speculations and possible directions for future research on Toxoplasma's effect on organizational dynamics.

Toxoplasma gondii (TOXO) is a neuroinvasive protozoan parasite that induces the formation of persistent cysts in mammalian brains. It infects approximately 1.1 million people in the United States annually. Latent TOXO infection is implicated in the etiology of psychiatric disorders, especially schizophrenia (SCZ), and has been correlated with modestly impaired cognition. The acoustic startle response (ASR) is a reflex seen in all mammals. It is mediated by a simple subcortical circuit, and provides an indicator of neural function. We previously reported the association of TOXO with slowed acoustic startle latency, an index of neural processing speed, in a sample of schizophrenia and healthy control subjects. The alterations in neurobiology with TOXO latent infection may not be specific to schizophrenia. Therefore we examined TOXO in relation to acoustic startle in an urban, predominately African American, population with mixed psychiatric diagnoses, and healthy controls. Physiological and diagnostic data along with blood samples were collected from 364 outpatients treated at an inner-city hospital. TOXO status was determined with an ELISA assay for TOXO-specific IgG. A discrete titer was calculated based on standard cut-points as an indicator of seropositivity, and the TOXO-specific IgG concentration served as serointensity. A series of linear regression models were used to assess the association of TOXO seropositivity and serointensity with ASR magnitude and latency in models adjusting for demographics and psychiatric diagnoses (PTSD, major depression, schizophrenia, psychosis, substance abuse). ASR magnitude was 11.5% higher in TOXO seropositive subjects compared to seronegative individuals (p = 0.01). This effect was more pronounced in models with TOXO serointensity that adjusted for sociodemographic covariates (F = 7.41, p = 0.0068; F = 10.05, p = 0.0017), and remained significant when psychiatric diagnoses were stepped into the models. TOXO showed no association with startle latency (t = 0.49, p = 0.63) in an unadjusted model, nor was TOXO associated with latency in models that included demographic factors. After stepping in individual psychiatric disorders, we found a significant association of latency with a diagnosis of PTSD (F = 5.15, p = 0.024), but no other psychiatric diagnoses, such that subjects with PTSD had longer startle latency. The mechanism by which TOXO infection is associated with high startle magnitude is not known, but possible mechanisms include TOXO cyst burden in the brain, parasite recrudescence, or molecular mimicry of a host epitope by TOXO. Future studies will focus on the neurobiology underlying the effects of latent TOXO infection as a potential inroad to the development of novel treatment targets for psychiatric disease.

here is evidence that some maternal infections during the prenatal period are associated with neurodevelopmental disorders, such as childhood autism. However, the association between autism and Toxoplasma gondii (T. gondii), an intracellular parasite, remains unclear. The authors examined whether serologically confirmed maternal antibodies to T. gondii are associated with odds of childhood autism in offspring. The study is based on a nested case-control design of a large national birth cohort (N=1.2 million) and the national psychiatric registries in Finland. There were 874 cases of childhood autism and controls matched 1: 1 on date of birth, sex, birthplace and residence in Finland. Maternal sera were prospectively assayed from a national biobank for T. gondii IgM and IgG antibodies; IgG avidity analyses were also performed. High maternal T. gondii IgM antibody was associated with a significantly decreased odds of childhood autism. Low maternal T. gondii IgG antibody was associated with increased offspring odds of autism. In women with high T. gondii IgM antibodies, the IgG avidity was high for both cases and controls, with the exception of three controls. The findings suggest that the relationship between maternal T. gondii antibodies and odds of childhood autism may be related to the immune response to this pathogen or the overall activation of the immune system.

Autoantibodies that bind the N-methyl-D-aspartate receptor (NMDAR) may underlie glutamate receptor hypofunction and related cognitive impairment found in schizophrenia. Exposure to neurotropic pathogens can foster an autoimmune-prone environment and drive systemic inflammation leading to endothelial barrier defects. In mouse model cohorts, we demonstrate that infection with the protozoan parasite, Toxoplasma gondii, caused sustained elevations of IgG class antibodies to the NMDAR in conjunction with compromised blood-gut and blood-brain barriers. In human cohorts, NMDAR IgG and markers of barrier permeability were significantly associated with T. gondii exposure in schizophrenia compared with controls and independently of antipsychotic medication. Combined T. gondii and NMDAR antibody seropositivity in schizophrenia resulted in higher degrees of cognitive impairment as measured by tests of delayed memory. These data underscore the necessity of disentangling the heterogeneous pathophysiology of schizophrenia so that relevant subsets eligible for NMDAR-related treatment can be identified. Our data aid to reconcile conflicting reports regarding a role of pathological NMDAR autoantibodies in this disorder.

Background. Human immunodeficiency virus (HIV)-associated neurocognitive disorders persist despite suppressive antiretroviral therapy (ART). Because latent Toxoplasma infection (LTI) may adversely impact brain function, we investigated its impact on neurocognitive impairment (NCI) in people living with HIV disease. Methods. Two hundred sixty-three HIV-infected adults underwent comprehensive neurocognitive assessments and had anti-Toxoplasma gondii immunoglobulin G (anti-Toxo IgG) measured by qualitative and quantitative enzyme-linked immunosorbent assays. Results. Participants were mostly middle-aged white men who were taking ART (70%). LTI was detected in 30 (11.4%) participants and was associated with a significantly greater prevalence of global NCI (LTI positive [ LTI+]=57% and LTI negative [ LTI-]=34%) (odds ratio, 1.67; 95% confidence interval, 1.17-2.40; P=.017). Deficits were more prevalent in the LTI+ vs the LTI -group in 6 of 7 cognitive domains with statistical significance reached for delayed recall (P<.01). The probability of NCI increased with higher CD4(+) T-cell counts among LTI+ individuals but with lower CD4(+) T-cell counts in LTI-persons. A strong correlation (r=.93) between anti-Toxo IgG levels and global deficit score was found in a subgroup of 9 patients. Biomarkers indicative of central nervous system inflammation did not differ between LTI+ and LTI-participants. Conclusions. In this cross-sectional analysis, LTI was associated with NCI, especially in those with higher CD4(+) T-cell counts. Longitudinal studies to investigate the role of neuroinflammation and neuronal injury in LTI patients with NCI and trials of anti-Toxoplasma therapy should be pursued.

Association between anti-Toxoplasma gondii antibodies and schizophrenia and psychotic bipolar in patients hospitalized in the psychiatric ward

Background Toxoplasmosis is caused by an intracellular parasite and is a worldwide disease. In laboratory, the parasites that cause the disease increases levels of dopamine in the brain tissue of treated mice. The evidence showed that dopamine releasing in the nucleus accumbens by activating the retro hippocampal region can disrupt the fornix section of brain as evolve to develop a psychosis in human. Methods This retrospective case-control study was conducted in patients with schizophrenia and psychotic bipolar referred to psychiatric clinic in Amir Kabir hospital, Arak. After psychiatric diagnosis of patients with schizophrenia and bipolar disorder, 2ml of blood samples were acquired from 76 patients and 75 controls without any psychotic illness or bipolar disorder and other mental illnesses with safety issues. The serum of samples was separated in laboratory and was kept until the time of testing at -20 degrees C. After collecting all the samples; Anti-Toxoplasma IgG on the case and control samples were analysed by ELISA. Results were analysed by SPSS software version 16 and were calculated by chi 2 tests. Results 55.26 per cent of patients (42 persons) were infected with Toxoplasma gondii and in the control group 36 per cent (27 patients) were infected, that the different between them was significant (p<0/05). Toxoplasmosis in psychotic men was more than psychotic women significantly (p<0/05). Toxoplasma gondii infection in patients with schizophrenia with psychotic bipolar patients showed no significant difference. Conclusion Patients with schizophrenia and bipolar disorder showed significantly higher rate than healthy people against Toxoplasma gondii infection. Whether prevention of infection with these parasites is effective in reducing the risk of psychosis requires further investigation.

Latent toxoplasmosis is associated with neurocognitive impairment in young adults with and without chronic HIV infection

We evaluated the impact of latent toxoplasmosis (LT) on neurocognitive (NC) and neurobehavioural functioning in young adults with and without chronic HIV infection, using a standardised NC test battery, self-reported Beck Depression Inventory, Frontal System Behavior Scale, MINI-International Neuropsychiatric Interview and risk-assessment battery. 194 young adults (median age 24 years, 48.2% males) with chronic HIV infection (HIV +) since childhood and 51 HIV seronegative (HIV) participants were included. HIV + individuals had good current immunological status (median CD4: 479 cells/mu l) despite a low CD4 nadir (median: 93 cells/mu l). LT (positive anti-Toxoplasma IgG antibodies) was present in one third of participants. The impairment rates in the HIV with and without Toxo were not significantly different (p = 0.17). However, we observed an increasing trend (p < 0.001) in impairment rates with HIV and LT status: HIV -/LT- (6.1%); HIV -/LT+ (22%), HIV +/LT- (31%), HIV +/LT+ (49%). In a multi variable analysis using the entire study group there were main effects on cognition for HIV and also for LT. Within the HIV+ group LT was associated with worse performance globally (p = 0.006), in memory (p = 0.009), speed of information processing (p = 0.01), verbal (p = 0.02) and learning (p = 0.02) domains. LT was not associated with depressive symptoms, frontal systems dysfunction or risk behaviors in any of the groups. HIV participants with lower Toxoplasma antibody concentration had worse NC performance, with higher GDS values (p = 0.03) and worse learning (p = 0.002), memory (p = 0.006), speed of information processing (p = 0.01) T scores. Latent Toxoplasmosis may contribute to NC impairment in young adults, including those with and without chronic HIV infection.

Reduced ERPs and theta oscillations underlie working memory deficits in Toxoplasma gondii infected seniors

Toxoplasma gondii is one of the most widespread infections in humans. Recent studies give evidence for memory deficits in infected older adults. To investigate working memory dysfunction in infected elderly, a double-blinded electrophysiological study was conducted. 84 persons derived from a sample of 131 healthy participants with the mean age of 70 years were assigned to two groups of 42 non-infected and 42 infected individuals. The outcome measures were behavioral performance, target and response-related ERPs, and time-frequency wavelets during performance in a n-back working-memory task. The infected individuals showed a reduced rate of detected targets and diminished P3b amplitude both in target-locked as well as response-locked data compared to the non-infected group. Time-frequency decomposition of the EEG-signals revealed lower evoked power in the theta frequency range in the target-locked as well as in the response-locked data in infected individuals. The reported effects were comparable with differences between healthy young and old adults described previously. Taking together, the reduced working-memory performance accompanied by an attenuated P3b and frontal theta activity may suggest neurotransmitter imbalance like dopamine and norepinephrine in T. gondii infected individuals. In face of a high prevalence of T. gondii infection and the increasing ratio of older population their accelerated memory decline may have substantial socioeconomic consequences.

Possible link between Toxoplasma gondii infection and mood disorders in Lorestan Province, Western Iran

Background: A large number of epidemiological evidence in humans and experimental studies in rodents have hypothesized that Toxoplasma gondii is a potentially relevant etiological factor in some mood disorders. Objectives: The aim of this study was to investigate whether latent toxoplasmosis has any role in mood disorders such as schizophrenia and bipolar disease in Western Iran. Patients and Methods: A total of 170 patients including 85 schizophrenia and 85 bipolar patients referred to the only psychiatric hospital in Lorestan province in Khorramabd city, Western Iran, and 170 healthy volunteers were screened for IgG and IgM anti-T. gondii antibodies by the Enzyme Linked Immunosorbent Assay (ELISA). Structured questionnaires were used to obtain information on risk factors for T. gondii infection. Results: Out of the 170 psychiatric patients, 103 (63.5%) patients were seropositive for IgG antibody and 14 (8.2%) were seropositive for IgM antibody. Of the 85 bipolar patients, 54 (63.5%) and 6 (7%) were found positive for anti-T. gondii IgG and IgM antibodies, respectively; whereas among 85 schizophrenia patients, 49 (57.6%) and 8 (9.4%) samples were found positive for anti-T. gondii IgG and IgM antibodies, respectively. From 170 healthy volunteers, 73 (42.9%) tested seropositive for anti-T. gondii antibodies; 65 (38.2%) tested seropositive for IgG antibody and 8 (4.7%) tested seropositive for IgM antibody. There was a significant difference in T. gondii IgG positivity between the psychiatric patients and control groups (P = 0.009). Several risk factors, which were significantly related to T. gondii seropositivity in psychiatric patients are being in contact with cats (P = 0.004), eating unwashed raw vegetables (P < 0.001), raw-milk/egg consumption (P = 0.001), and drinking clean water (P = 0.035). Conclusions: We concluded that psychiatric patients in general and schizophrenia and bipolar cases in particular had a significantly higher relative frequency of T. gondii infection than healthy individuals of Lorestan Province, Western Iran.

Toxoplasma gondii is a ubiquitous protozoan parasite with approximately one-third of the worlds' population chronically infected. In chronically infected individuals, the parasite resides primarily in cysts within neurons in the central nervous system. The chronic infection in immunocompetent individuals has been considered to be asymptomatic but increasing evidence indicates the chronic infection can lead to neuropsychiatric disorders such as Schizophrenia, prenatal depression and suicidal thoughts. A better understanding of the mechanism(s) by which the parasite exerts effects on human behavior is limited due to lack of suitable human neuronal models. In this paper, we report the use of human neurons derived from normal cord blood CD34+ cells generated via genetic reprogramming, as an in vitro model for the study T. gondii in neurons. This culture method resulted in a relatively pure monolayer of induced human neuronal-like cells that stained positive for neuronal markers, MAP2, NFL, NFH and NeuN. These induced human neuronal-like cells (iHNs) were efficiently infected by the Prugniad strain of the parasite and supported replication of the tachyzoite stage and development of the cyst stage. Infected iHNs could be maintained through 5 days of infection, allowing for formation of large cysts. This induced human neuronal model represents a novel culture method to study both tachyzoite and bradyzoite stages of T. gondii in human neurons.

Prior research has suggested an association between exposure to infectious disease and neurocognitive function in humans. While most of these studies have explored individual viral, bacterial, and even parasitic sources of infection, few have considered the potential neurocognitive burden associated with multiple infections. In this study, we utilized publically available data from a large dataset produced by the Centers for Disease Control and Prevention that included measures of neurocognitive function, sociodemographic variables, and serum antibody data for several infectious diseases. Specifically, immunoglobulin G antibodies for toxocariasis, toxoplasmosis, hepatitis A, hepatitis B, and hepatitis C, cytomegalovirus, and herpes 1 and 2 were available in 5662 subjects. We calculated an overall index of infectious-disease burden to determine if an aggregate measure of exposure to infectious disease would be associated with neurocognitive function in adults aged 20-59 years. The index predicted processing speed and learning and memory but not reaction time after controlling for age, sex, race-ethnicity, immigration status, education, and the poverty-to-income ratio. Interactions between the infectious-disease index and some sociodemographic variables were also associated with neurocognitive function. In summary, an index aggregating exposure to several infectious diseases was associated with neurocognitive function in young- to middle-aged adults. (C) 2015 Elsevier Inc. All rights reserved.

The prevalence of anti-Toxoplasma gondii antibodies in stutterers is higher than in the control group

The purpose of this study was to investigate the possible relationship between Toxoplasma gondii (Nicolle et Manceaux, 1908) and stuttering. We investigated the seropositivity rate for anti-T. gondii IgG and antibodies by enzyme-linked immunosorbent assay (ELISA) in stuttering children to ascertain a possible relationship between T. gondii infection and stuttering. We selected 65 stuttering children and 65 control children (non-stutterers) to investigate the seropositivity rate of anti-T. gondii antibodies by ELISA. Cranial magnetic resonance imaging (MRI) and scalp electroencephalography (EEG) were also performed in stuttering children. The seropositivity rate of anti-T. gondii IgG antibodies among stuttering children (28%) was significantly higher than in control group (5%; p = 0.001). No abnormality was detected in cranial MRI's of stutttering children and their EEG recordings were also normal. There was no significant difference in seropositivity rate regarding age, genders and residence area. The association between seroprevalence of infection with T. gondii and stuttering may be due to hyperdopaminergic state in brains of patients who are T. gondii-seropositive. Thus, there might be a causal relationship between toxoplasmosis and stuttering.

Influence of Toxoplasma gondii infection on male fertility: A pilot study on immunocompetent human volunteers

Background: This study was conducted to investigate the influence of Toxoplasma gondii infection on spermatic and hormonal parameters in a pilot sample of immunocompetent human male subjects. Methods: This cross sectional, observational pilot study on 60 immunocompetent human male subjects aged between 18 and 60 yr old was conducted between 2012-2013. Blind evaluation of serological markers of past T. gondii infection (TOX-IgG, TOX-IgM) was performed, along with individual spermiograms and determinations of follicle-stimulating hormone (FSH) and testosterone serum levels. Results: The overall prevalence of past T. gondii infection in the investigated immunocompetent male subjects was 25%. No statistically significant influence of T. gondii infection on sperm characteristics (ejaculate quantity, sperm count, motility, morphology) and serum levels of FSH or testosterone were found. Among possible predictors of a modified spermiogram studied by multiple logistic regression along with the T. gondii infection (age, smoking, alcohol consumption, fertility influencing malformations, infections, trauma or medication), only the presence of varicocele in the medical history of the studied subjects was found to significantly participate in the prediction of a modified spermiogram (P=0.0154). A necessary sample size of 994 subjects was computed in order to achieve a test power of 0.8 (80%) to discriminate an effect size of 8.89% estimated by our pilot study. Conclusions: Although our investigation did not demonstrate an influence of latent T. gondii infection on spermatic and hormonal parameters of immunocompetent male humans, the absence of such an influence cannot be affirmed, due to the limited sample size of our pilot study.

Background Toxoplasmosis is an important, widespread, parasitic infection caused by Toxoplasma gondii. The chronic infection in immunocompetent patients, usually considered as asymptomatic, is now suspected to be a risk factor for various neurological disorders, including epilepsy. We aimed to conduct a systematic review and meta-analysis of the available literature to estimate the risk of epilepsy due to toxoplasmosis. Methods A systematic literature search was conducted of several databases and journals to identify studies published in English or French, without date restriction, which looked at toxoplasmosis (as exposure) and epilepsy (as disease) and met certain other inclusion criteria. The search was based on keywords and suitable combinations in English and French. Fixed and random effects models were used to determine odds ratios, and statistical significance was set at 5.0%. Principal findings Six studies were identified, with an estimated total of 2888 subjects, of whom 1280 had epilepsy (477 positive for toxoplasmosis) and 1608 did not (503 positive for toxoplasmosis). The common odds ratio (calculated) by random effects model was 2.25 (95% CI 1.27-3.9), p = 0.005. Conclusions Despite the limited number of studies, and a lack of high-quality data, toxoplasmosis should continue to be regarded as an epilepsy risk factor. More and better studies are needed to determine the real impact of this parasite on the occurrence of epilepsy

ObjectiveTo perform a meta-analysis on studies reporting prevalence of Toxoplasma gondii (T.gondii) infection in any psychiatric disorder compared with healthy controls. Our secondary objective was to analyze factors possibly moderating heterogeneity. MethodA systematic search was performed to identify studies into T.gondii infection for all major psychiatric disorders versus healthy controls. Methodological quality, publication bias, and possible moderators were assessed. ResultsA total of 2866 citations were retrieved and 50 studies finally included. Significant odds ratios (ORs) with IgG antibodies were found in schizophrenia (OR 1.81, P<0.00001), bipolar disorder (OR 1.52, P=0.02), obsessive-compulsive disorder (OR 3.4, P<0.001), and addiction (OR 1.91, P<0.00001), but not for major depression (OR 1.21, P=0.28). Exploration of the association between T.gondii and schizophrenia yielded a significant effect of seropositivity before onset and serointensity, but not IgM antibodies or gender. The amplitude of the OR was influenced by region and general seroprevalence. Moderators together accounted for 56% of the observed variance in study effects. After controlling for publication bias, the adjusted OR (1.43) in schizophrenia remained significant. ConclusionThese findings suggest that T.gondii infection is associated with several psychiatric disorders and that in schizophrenia reactivation of latent T.gondii infection may occur.

Background Past research linked Toxoplasma gondii (TG) infection in humans with neurological and mental disorders (e.g., schizophrenia, Alzheimer's disease and attention disorders), irregularities of the dopaminergic and testosterone system, and increased likelihood of being involved in traffic accidents. Methodology/Principal Findings We test for an association between TG infection and financial decision-making (DM) using a case-control design in a sample of female Czech students (n = 79). We estimate each subject's risk attitude and loss aversion using an experimental economic task involving real monetary incentives. We find no significant evidence that either measure of decision-making is associated with TG infection. Conclusion We were unable to find evidence of an association between TG infection and financial decision- making in females.

The parasite Toxoplasma gondii has been found to manipulate the behavior of its secondary hosts to increase its own dissemination which is commonly believed to be to the detriment of the host (manipulation hypothesis). The manipulation correlates with an up-regulation of dopaminergic neurotransmission. In humans, different pathologies have been associated with T. gondii infections but most latently infected humans do not seem to display overt impairments. Since a dopamine plus does not necessarily bear exclusively negative consequences in humans, we investigated potential positive consequences of latent toxoplasmosis (and the presumed boosting of dopaminergic neurotransmission) on human cognition and behavior. For this purpose, we focused on action cascading which has been shown to be modulated by dopamine. Based on behavioral and neurophysiological (EEG) data obtained by means of a stop-change paradigm, we were able to demonstrate that healthy young humans can actually benefit from latent T. gondii infection as regards their performance in this task (as indicated by faster response times and a smaller P3 component). The data shows that a latent infection which is assumed to affect the dopaminergic system can lead to paradoxical improvements of cognitive control processes in humans.

Effects of latent toxoplasmosis on autoimmune thyroid diseases in pregnancy

Background: Toxoplasmosis, one of the most common zoonotic diseases worldwide, can induce various hormonal and behavioural alterations in infected hosts, and its most common form, latent toxoplasmosis, influences the course of pregnancy. Autoimmune thyroid diseases (AITD) belong to the well-defined risk factors for adverse pregnancy outcomes. The aim of this study was to investigate whether there is a link between latent toxoplasmosis and maternal AITD in pregnancy. Methods: Cross-sectional study in 1248 consecutive pregnant women in the 9–12 th gestational weeks. Serum thyroid- stimulating hormone (TSH), thyroperoxidase antibodies (TPOAb), and free thyroxine (FT4) were assessed by chemiluminescence; the Toxoplasma status was detected by the complement fixation test (CFT) and anti- Toxoplasma IgG enzyme-linked immunosorbent assay (ELISA). Results: Overall, 22.5% of the women were positive for latent toxoplasmosis and 14.7% were screened positive for AITD. Women with latent toxoplasmosis had more often highly elevated TPOAb than the Toxoplasma -negative ones ( p = 0.004), and latent toxoplasmosis was associated with decrease in serum TSH levels ( p = 0.049). Moreover, we found a positive correlation between FT4 and the index of positivity for anti- Toxoplasm a IgG antibodies ( p = 0.033), which was even stronger in the TPOAb-positive Toxoplasma -positive women, ( p = 0.014), as well as a positive correlation between FT4 and log 2 CFT ( p = 0.009). Conclusions: Latent toxoplasmosis was associated with a mild increase in thyroid hormone production in pregnancy. The observed Toxoplasma -associated changes in the parameters of AITD are mild and do not seem to be clinically relevant; however, they could provide new clues to the complex pathogenesis of autoimmune thyroid diseases

Background: This study investigated the possible role of toxoplasmosis in increasing the risk of traffic accidents. Materials and Methods: This unmatched casecontrol study was done on a total of 103 people (102 men and one woman) involved in a traffic accident as drivers for a time period of one year during 2011–2012.The controls were consisted of 200 people (88 men and 112 women), residents of the same region and in same age group. Blood samples were driven and all serum samples were separated and stored at -20 ºC until the testing. All collected sera were examined using ELISA technique. Results: For IgG seropositivity, a 2.02-fold increase (χ2 = 7.84, P = 0.004, 95%CI: 1.23 – 3.33) in risk for traffic accident was estimated. Furthermore, IgM seropositivity could not significantly increase the risk of traffic accident, with an OR of 0.73 (χ2 = 0.209, P = 0.461, 95%CI: 0.19 – 2.81). However, stratification analysis showed that only the individuals with age range ≤ 30-year were significantly more common in traffic accident than in controls (OR: 2.15, χ2 = 6.91, P = 0.007, 95%CI: 1.20 – 3.85). There was no statistically significant difference between the IgM and IgG seropositivity in case and control groups (X2 = 3.65, P = 0.09, 95%CI: 0.99 – 2.67). Conclusion: There is an increased risk for traffic accidents for drivers owing to these high seroprevalence of latent toxoplasmosis. Latent toxoplasmosis in drivers should be taken into account while developing preventive strategies for traffic accidents in Iran.

Effect of toxoplasmosis on personality profiles of Iranian men and women

There is evidence to suggest that the protozoan Toxoplasma gondii, which causes toxoplasmosis, changes the personality of people who are infected with it. The aim of this study was to compare the personality characteristics of Iranian students with and without latent toxoplasmosis. A total of 237 students (111 men and 126 women) of Jundishapur University of Medical Sciences (Ahvaz, Iran) were tested for the presence of anti-Toxoplasma antibodies and completed demographic questionnaires and Cattell's 16 Personality Factor Questionnaire. Data were analysed using multiple univariate analyses of variance. Women with latent toxoplasmosis had a significantly different personality profile from women without toxoplasmosis, namely higher O (apprehension), N (privateness) and Q(4) (tension) scores, and lower Q(1) (openness to change) scores. Infected men had significantly higher L (vigilance, mistrust) scores compared to non-infected men. Factors E (dominance) and Q(1) (openness to change) tended to be higher in infected men than non-infected men but the difference was not quite statistically significant. Our findings have, for the first time, independently confirmed that personality profile is affected by latent toxoplasmosis.

Association between seropositivity for Toxoplasma gondii, scholastic development of children and risk factors for T-gondii infection.

We evaluated the possible association of seropositivity for Toxoplasma gondii and certain risk factors for T. gondii infection with the scholastic development of children. One hundred children aged 613 years attending the Hospital Municipal de Maring Parana, Brazil, participated in the study. Serologic tests for IgG and IgM anti-T. gondii (indirect immunofluorescence (capture ELISA) were performed. The Scholastic Performance Test (SPT) for writing, mathematics and reading was applied to each child, and the result was classified as high, average or poor. The guardian of each child responded to a questionnaire about certain aspects of the childs living situation and diet. The prevalence of seropositivity for T. gondii was 8. An association between seropositivity for T. gondii and scholastic development in the mathematics subtest and also consumption of fresh cheese were observed. Children with exposed soil, sand or grass lawn in their peridomicile were 9.116 times more likely to be infected by T. gondii. The findings showed the need to test school-age children for this parasite, educate families with T. gondii-positive children, provide training to educators, monitor recreation areas, and raise awareness of the need for care in handling food.

Knowledge about the influence of latent toxoplasmosis on development and general biological condition of children is scant and thus the aim of the present study was to investigate these aspects in some detail. We compare school children in rural area seropositive and seronegative to the apicomplexan parasite Toxoplasma gondii (Nicolle et Manceaux, 1908) in terms of their developmental age, body mass and body height, physical fitness and end-of-term grades. Additionally, we evaluated the risk factors of infection with T gondii such as the presence of cats in the household and eating raw meat products. With IFAT and ELISA tests, the prevalence of anti-Toxoplasma antibodies was 41% (190 children examined) and the rate of infection was higher in girls (44%) than in boys (36%). No significant differences were observed in morphological features and physical fitness of examined children. In girls the level of developmental age measured with electrophoretical mobility of nuclei method was significantly higher and school performance significantly lower for those infected with Toxoplasma than for uninfected; this finding was particularly intriguing. Only boys who ate raw meat products were more likely to be seropositive. The study provides some new information on gender differences in reaction to Toxoplasma infection.

Objectives. To address the role of latent T. gondii infection in schizophrenia we studied the infl uence of latent toxoplasmosis on brain morphology. Methods. An optimized voxel-based morphometry of magnetic resonance imaging was analyzed by analysis of variance with diagnosis and seropositivity as factors in 44 schizophrenic patients (12 T. gondii positive) and 56 controls (13 T. gondii positive). Results. Grey matter (GM) volume was reduced in schizophrenia patients compared with controls in the cortical regions, hippocampus and in the caudate. In the schizophrenia sample we found a signifi cant reduction of GM volume in T. gondii positive comparing with T. gondii -negative patients bilaterally in the caudate, median cingulate, thalamus and occipital cortex and in the left cerebellar hemispheres. T. gondii -positive and -negative controls did not differ in any cluster. Among participants seropositive to T. gondii the reduction of GM in the schizophrenia subjects was located in the same regions when comparing the entire sample (11,660 over-threshold voxels ( P  0.05, FWR corrected). The differences between T. gondii -negative patients and controls consisted only of 289 voxels in temporal regions. Conclusions. Our study is the fi rst to document that latent toxoplasmosis reduces GM in schizophrenia but not in controls.

Cognitive deficits are a core feature of schizophrenia. Epidemiological evidence indicates that microbial pathogens may contribute to cognitive impairment in patients with schizophrenia. Exposure to Toxoplasma gondii (T. gondii) has been associated with cognitive deficits in humans. However, the mechanisms whereby the parasite impacts cognition remain poorly understood. Animal models of T. gondii infection may aid in elucidating the underpinnings of cognitive dysfunction. Here, we (1) overview the literature on the association of T. gondii infection and cognitive impairment, (2) critically analyze current rodent models of cognitive deficits resulting from T. gondii infection, and (3) explore possible mechanisms whereby the parasite may affect cognitive function.

Brain cancer mortality rates increase with Toxoplasma gondii seroprevalence in France

The incidence of adult brain cancer was previously shown to be higher in countries where the parasite Toxoplasma gondii is common, suggesting that this brain protozoan could potentially increase the risk of tumor formation. Using countries as replicates has, however, several potential confounding factors, particularly because detection rates vary with country wealth. Using an independent dataset entirely within France, we further establish the significance of the association between T. gondii and brain cancer and find additional demographic resolution. In adult age classes 55 years and older, regional mortality rates due to brain cancer correlated positively with the local seroprevalence of T. gondii. This effect was particularly strong for men. While this novel evidence of a significant statistical association between T. gondii infection and brain cancer does not demonstrate causation, these results suggest that investigations at the scale of the individual are merited.
